What is

Tesamorelin

Tesamorelin is a synthetic peptide hormone used to promote weight loss and reduce the accumulation of abdominal fat. It works by increasing the production of growth hormone in the body, which has been shown to help reduce the amount of fat stored in the abdominal region.

Tesamorelin acts as a potent agonist of the growth hormone-releasing hormone (GHRH) receptor, which is found in the pituitary gland. The pituitary gland is a small but crucial endocrine gland located at the base of the brain, and is responsible for the regulation of several hormones in the body, including growth hormone. By stimulating the GHRH receptor, tesamorelin increases the production of growth hormone in the body, leading to a reduction in the amount of abdominal fat stored.

In clinical studies, tesamorelin has been shown to be effective in reducing abdominal fat and improving the body’s shape in individuals with HIV-associated lipodystrophy. However, its use for weight loss and reduction of abdominal fat has also been investigated in other populations, including those without HIV.

Protocols

Provided as a 10mg lyophilized vial

  • Initiation: Tesamorelin therapy is initiated to enhance growth hormone secretion and support various metabolic processes.
  • Initial Dose: Begin with 0.5 mg (0.05 ml or 5 units) subcutaneously 5 days per week (5/7 days) for the first two weeks, administered while fasting 2-3 hours prior to injection.
  • Titration: If well tolerated, increase the dose to 1 mg (0.1 ml or 10 units) subcutaneously 5 days per week (5/7 days) after the initial two-week period.
  • Maintenance: Maintain the dose at 1 mg (0.1 ml or 10 units) subcutaneously 5 days per week for ongoing support in growth hormone secretion and metabolic enhancement.
  • Duration:Tesamorelin is administered five days per week.
  • Weeks 1-2: 0.5 mg (0.05 ml or 5 units) of Tesamorelin 5 days per week
  • Week 3 and onwards: 1 mg (0.1 ml or 10 units) of Tesamorelin 5 days per week

*The dosage of Tesamorelin may be adjusted based on the individual’s response, medical history, and specific treatment goals.

  • Known hypersensitivity to Tesamorelin or any component of the formulation.
  • Not recommended for individuals with active malignancy, pregnancy, or breastfeeding.
  • Caution in patients with pituitary gland issues or diabetes.

  • Regular monitoring of growth hormone levels and metabolic parameters to assess the effectiveness of the treatment.
  • Periodic blood tests to check for any adverse effects and to adjust the treatment as necessary.

Procedure

Reconstitution

  • Draw 1 ml of sterile water into a syringe.
  • Inject the sterile water into a vial containing 10 mg of Tesamorelin peptide.
  • Gently swirl the vial until the peptide is fully dissolved.

  • Remove the cap from a tuberculin syringe.
  • Draw the reconstituted Tesamorelin solution into the syringe to the required dosage (0.05 ml or 5 units initially, 0.1 ml or 10 units after titration).
  • Clean the selected injection site with an alcohol swab.
  • Pinch the sanitized skin and insert the needle at a 90-degree angle.
  • Administer the injection subcutaneously.
  • Administer while fasting, 2-3 hours prior to injection.

  • Store the remaining Tesamorelin solution in the refrigerator at 2-8°C.
  • Before the next use, clean the vial top with an alcohol swab.
  • Use a new syringe for each administration.
  • Ensure to rotate injection sites (abdomen, thigh, or upper arm) to minimize skin irritation.

Results:

Tesamorelin, a synthetic analog of growth hormone-releasing hormone (GHRH), has demonstrated significant potential in promoting weight loss, particularly by reducing visceral adipose tissue (VAT). Clinical trials have shown that Tesamorelin effectively targets and reduces VAT, which is a major contributor to obesity-related health issues.

In a randomized, placebo-controlled trial involving patients with type 2 diabetes, Tesamorelin treatment resulted in significant reductions in body weight and VAT. Participants experienced improvements in body composition, including reductions in waist circumference and body fat percentage. Additionally, Tesamorelin improved metabolic parameters such as insulin sensitivity, fasting glucose, and lipid profiles (total cholesterol, LDL cholesterol, HDL cholesterol, and triglycerides).

The trial also highlighted Tesamorelin’s favorable safety profile, with most adverse effects being mild and transient, including injection site reactions and occasional muscle pain. Importantly, there were no serious adverse events, indicating that Tesamorelin is well-tolerated.
